Method Actions

Prime - clean valve and pump with selected buffer(s).

Buffer(s) are drawn through valves 1 and 2 into the pumps and then pushed out into the top waste instead of flowing through the columns. This can be used to fill or clean buffer tubing.

Condition – equilibrate or wash column with selected buffer(s).

Buffer(s) are pushed through the columns into waste or flowthrough. The difference between Condition and Collect is that Collect will push buffers into a Fraction.

Add Samp – load sample onto column.

Sample(s) are drawn and pushed through the columns and then into waste or flowthrough.

Reload - transfer flowthrough from collection vessel back to the sample bottles.

This step allows the flowthrough to the re-applied to the protein purification column without manual intervention.

Collect - elute with selected buffer(s) and collect eluent in specified fraction tubes.

Buffer(s) are pushed through the columns to elute analytes into Fraction 1 or Fraction 2 (up to 6 for 6-fraction systems). Samples can also be directly collected into Fraction 1 or Fraction 2.

Air-Purge - use air to purge residual fluid through columns into fractions/waste.

Outside air is drawn in and then pushed into sample containers (e.g., for pushing remaining rinsate into the sample bottle) or delivered through columns into fractions or waste.

Clean - Deliver selected buffer(s) into sample containers, optionally followed by an air purge to clear the lines.

This step may be used for cleaning or filling the sample lines. The system delivers selected buffer(s) to the sample containers.

Wait - pause run for a set time or until the user manually triggers the next step.

The “Time-based” option will pause the system for a set time. The “Manual resume” option will continue pausing until the operator taps “start” to resume. A loud “beep” is triggered for the “Manual Resume” option to get the operator’s attention. The “Manual resume” option can be used to allow operator intervention such as changing out fraction tubes.
